As far as I know sound quality will not be affected as long as the material you use is securely mounted. The only time sound quality is affected is in an enclosure of some type. I prefer lexan over plexiglass as it does not crack as easily. Actually it has a 10 year warranty against cracking. and it is only around $1.50 more per sq ft.

Agreed, Lexan is the best choice here for strength and durability over plexiglass. A 1/4" -3/8" thickness is adequate. You can also use wood mounting plates which are easier to cut (tooling) and much cheaper too. Can always paint the wood to hide it from view to a degree... through the grill cover plates.

Lexan is tough to tool (cut) as it will melt at high cutting speeds of a Router, Dremel or Roto Zip. You'd have to practice cutting Lexan a bit to figure out the right cutting tool and feed speed. A circle cutting jig or compass tool adapter is fine for 4" diameter holes or larger. A hole cutting tool on a drill will do smaller holes well... but watch your speed/pressure as you drill. If the material get too hot, it melts and may distort the cut.

use mdf. A router will allow you to make identical and very nice baffles if you know how to used it. A jig saw wil do just as well. If you use 1/2" you can fit a 6.5 easily
