I have two 12in 1000w subs in a ported box. I have a 2000w 2ch amp. I was wanting to put a neon tube in the back of my box so it would shine through the port. Is there a chace of the tube shattering?

Most neons I have seen, are pretty sturdy in construction. If they had standoff mounts with sound absorbent sleeves to hold them, should be fine.
If they can be mounted under the car, they can handle the force most subs can produce.

they should be fine. You could also use led's if you want it brighter.
